To efficiently block of the activity of Calpain or NOS, bath application of the calpain activity inhibitor Calpeptin (250 μM, C8999, Sigma; dissolved in DMSO) (Tsujinaka et al., 1988) or the NOS inhibitor NG-Monomethyl-L-arginine (L-NMMA, 500 μM, M7033, Sigma; dissolved in water) (Kashiwagi et al., 2005; Rees et al., 1989) was used. A cut of the skin (~50 μm in the length) between the two mesencephalic veins (MsVs) at the dorsal brain was made with a micropipette.

2019, Biochemical and Biophysical Research CommunicationsCitation Excerpt :When cytosolic free Ca2+ was elevated using ionomycin, after a delay of about 100sec, the rate of production was significantly increased (rate constant 1.48 × 103s−1 to 5.77 × 103 s−1; Fig. 1b). After the Ca2+ activated increase in R110 production was triggered, the addition of the calpain inhibitor, calpeptin [15] inhibited the further rise, pointing to calpain as the source of the R110 increase. Interestingly, calpeptin also inhibited the “non-activated “production of R100 and there was no significant production after calpain inhibition in ionomycin treated cells (Fig. 1c), suggesting that all increases in R110 were the result of calpain activity.
